West Ham are set to leave out several first-team regulars for their second-leg trip to Romanian side Astra Giurgiu with their tie level at 2-2.
But Koeman will not do the same at Vitesse Arnhem on Thursday despite holding a 3-0 lead from the first leg.
"It is a crazy thing to think about it, to throw everything away in the start of the Europa League legs," he said.
"I am a person who likes to win always and I hate losing. I hate to lose and that mentality needs my players as well."
Koeman will be forced into one change from the side that comfortably beat Vitesse at St Mary's last week, with midfielder Jordy Clasie - signed from Feyenoord for £8m in July - ruled out with a hamstring injury.
Saints' first Premier League match of the season at Newcastle on Saturday starts fewer than 65 hours after the final whistle in the Netherlands.
But Koeman believes wholesale changes could undermine the effort expended in securing seventh place last season.
"We did a great season last year and we played for the Europa League over 38 games in the Premier League," the Dutchman said.
"Football is sometimes strange and if you don't start with enough concentration, if you don't play with the same intensity in pressing the opponent, then it is possible that you get a difficult evening."
Striker Jay Rodriguez may return after 16 months out with a knee ligament injury, while defender Steven Caulker, on loan from QPR, could make his debut.
Southampton, Manchester United and Arsenal are reportedly keen on signing Celtic defender Virgil van Dijk.
"Everybody knows he's a good player - it's a lot of interest for the player. Let's see what happens," Koeman said.
Three players on loan from Chelsea - Isaiah Brown, Lewis Baker and Danilo Pantic - played for Vitesse in the first leg, while striker Dominic Solanke has since arrived on loan from Stamford Bridge.

If Spurs and Liverpool can't with all the fucking players they've both got; Southampton won't last...
Still, it's a shit competition anyway... It's only worthwhile if you get a big game against a top side and that's that...
Gone are the more simpler days of straight knock-out rounds without Group Stage bollocks...
I bet we only had to go through 7 or 8 rounds to win the ECWC in '65 and reach the final again in '76...

The last month of the season people were more bothered about how many yellow cards we got rather than how many goals.... if you gain qualification to any cup by being the best behaved team in the league then in my eyes that is a fraud qualification... if we had sweated blood sweat and tears and over achieved like Southampton did then maybe I would feel differently

Northern : Once you get there - it shouldn't matter how you got there - it should matter that you are there. We had the same chance as Southampton to progress and play in Europe this year. I don't agree that they have a better squad than us - I don't agree that they played harder because they won their way in - and we got in through the back door.
I do however agree that they have grasped their opportunity with both hands - while we played with both hands tied behind our back..... puzzling and bemusing to me.... Now I do worry whether Slavan knows what he is doing....
